Description: Independent transgenic line #2 containing the genetically encoded fluorescent biosensor Kinase Localization Reporter variant KLR-EE. KLR-EE consists of a bipartite modified nuclear localization signal (bNLS) with two phosphosites, a nuclear localization signal (NLS), the mNeonGreen fluorophore, a T2A "self-cleaving" Thosea asigna virus 2A peptide, the mRuby3 fluorophore, and an NLS derived from simian virus 40 (SV40). The two phosphosites within the bNLS have been mutagenized to phosphomimetic glutamic acids. This control construct does not contain a kinase docking domain. Primary transformants were identified with an epifluorescence microscope and seed was collected from plants exhibiting strong fluorescence. Transgene genotype unknown.
