Description: Quintuple mutant (met1-8, drm1-2, drm2-2, cmt3-11, cmt2-3) generated by CRISPR/Cas9-mediated mutation of MET1 directly in the ddcc quadruple mutant background. Two sgRNAs were designed to target the first exon of the MET1 gene and the CRISPR-Cas9 construct was transformed into the ddcc mutant background. Then, heterozygous met1 mutants (met1/+) without the CRISPR-Cas9 construct were isolated from the mutated T2 populations in the ddcc background. Deposited seeds are the progenies of met1/+ drm1 drm2 cmt3 cmt2 plants and were used to isolate mddcc DNA methylation-free quintuple mutant plants.

Quintuple homozygous plants exhibit extreme growth retardation and small size, display a suite of severe developmental defects, and do not undergo floral transition.
