Germination Medium (GM)

This medium is used for Arabidopsis seed germination. It is based on very old recipes (Knop, 1860) for hydroponic root culture and works very well for (almost natural) root development in agar.


Stock Solutions:  (use double distilled water for all solutions, stocks are stable at 4C)
					stock conc.				
1	KNO3				1.2 M		12 g / 100 ml 
	MgSO4.7H2O			80 mM		1.97 g / 100 ml
2	Ca(NO3)2.4H2O			30%		30 g / 100 ml	
3	KH2PO4				1 M		13.6 g / 100 ml	
4	FeNaEDTA			20 mM		0.73 g / 100 ml

To prepare 1 liter germination medium; add to distilled water:
2.5 	ml stock 1
1	ml stock 2
0.5	ml stock 3
0.5 	ml stock 4
0.25 	ml stock 5,
adjust pH to 6.4 with 1N KOH (ca. 125 microlitre / l medium)
add 6g agar / liter (preferably tissue culture quality)
