Description: Pt-GFP is a pH-reporter protein when used ratiometrically in the double excitation mode. It has a broad pH-responsiveness and a good acid stability.Arabidopsis lines PtGFP6-3 and PtGFP_Kn can be used to monitor cytoplasmic pH in a wide range of applications. PtGFP_kn and PtGFP6-3 represent two expression lines from two different floral dip experiments. Hence the T-DNA from the Agrobacs carrying the PtGFP gene has been inserted in different (as yet not identified) locations of the Arabidopsis genome, leading to different expression levels (kn appears a bit brighter than 6-3).

Phenotype
These lines express green fluorescent protein from the seapen Ptilosarcus gurneyi (Pt-GFP) constitutively in the cytoplasm under control of a single 35S-CaMV-promoter.Cells fluoresce green when exited with blue light.There are no other differences in phenotype neither in appearance and growth nor in development and reproduction when compared to Col-0 wild type.
