Description: ProCESA2::GUS, transgenic line containing the cellulose synthase 2 (CESA2) promoter, E. coli B-glucuronidase (GUS) reporter gene; genomic fragments upstream of the CESA initiating AUG codon were amplified by PCR, cloned into pRITA upstream of the GUS gene, and sequenced to confirm accuracy of amplification; the promoter:GUS cassettes were then cloned at NotI fragments into the Ti plasmid pMLBART in the same orientation as the BAR gene, introduced into Agrobacterium strain ASE1 by electroporation, and used to transform Col-0 Arabidopsis by the floral dip method; transgenic plants were selected with a 1:1,000 dilution of Finale (Hoechst-Roussel Agri-Vet, Somerville, NJ) twice a week for three weeks; expression of GUS was confirmed by the standard histological assay at several subsequent generations.
