Description: MAP65-1 genomic DNA was PCR amplified from Col-0 DNA and inserted into pPZP211. eGFP sequence was amplified without the stop codon and cloned upstream of MAP65-1 gene in pPZP211 vector. 2.6 kb upstream of MAP65-1 gene was inserted upstream of eGFP to drive gene expression. Agrobacterium strain GV3001 was used to transform map65-1-2 homozygous mutant line, derived from SALK_118225. T1 seeds were selected on Kanamycin. Plants are segregating for the transgene.
